| Product Details | 【产品简介】 蛋白分子量标准，常称为蛋白marker，是蛋白电泳及免疫印迹实验不可或缺的工具。蛋白marker不仅提示蛋白电泳是否正常进行，也能够简单判断转膜是否成功。彩色预染蛋白marker （8-180 kDa）是由10种纯化的蛋白混合而成，各条带浓度约为0.2-0.4 mg/ml。蛋白与染料共价结合，其中72 kDa条带为红色预染条带，8 kDa条带为绿色预染条带，方便判断各个条带的准确位置，条带清晰锐利，易于观测，适合蛋白电泳以及后续转膜。 【产品详情】 【使用方法】 1 将本产品于室温或则冰上融化后，轻柔混匀，使沉淀充分溶解（切勿煮沸加热）； 2 吸取3-5 μL到SDS-聚丙烯酰胺胶的上样孔中，电泳和转膜； 3 电泳结束后，通过蛋白染色液观察目的条带。 【注意事项】 1 本品为即用型产品，融化后直接吸取即可，使用前无需加热、还原或添加样品缓冲液。 2 每次吸取都要使用灭过菌的枪头或干净的针头，用完马上盖严，避免引入蛋白酶使产品降解。 3 建议分装保存，避免反复冻融。 4 转膜缓冲体系中SDS不要超过0.02-0.04%。 【常见问题】 问题1 Marker所有条带一定会都跑出来吗？ 回答1 不一定。实际电泳中，所见条带数比说明书上条带数少1-2条是有可能的，由于电泳时间不足、凝胶浓度偏低等因素造成。给出的建议是，目的蛋白附近的marker条带清晰可见即可，或者溴酚蓝前沿刚跑出胶的时候再停止电泳。 问题2 Marker的条带变宽怎么解决? 回答2 如果是上样量过多引起的，减少上样量即可；溶解不完全，在上样前充分溶解。 问题3 为什么有时候有些条带会消失? 回答3 凝胶浓度过低可能会导致小分子量条带分散，凝胶浓度过高则可能会导致高分子量条带分散。检査产品有效期和储存条件，失效的产品可能会不稳定，出现褪色或缺失条带。确保产品未经加热或煮沸。 问题4 预染蛋白marker可以用来准确预估蛋白分子量大小吗? 回答4 不可以。预染marker中每个蛋白质均与染料共价结合，这些染料可能会导致marker中蛋白在不同缓冲系统、不同凝胶中有相似但不相同的迁移速率。如需准确预估分子量，建议使用非预染marker。 |
